Depending on the type of membership site you are wanting to create - there are several options. Among my favorites are:
1. For basic sites, using Wordpress with Members Plugin will work - there are plenty out there ranging in price and functionality.
2. Something more functional and scaleable including user forums, downloads, etc, I would recommend using e107, a content management system with a growing developer base. Amember is a good way to get a membership site off the press quickly using e107
3. Semi-custom - find a platform you like (e107, Drupal, Joomla, Mambo, etc) get plugins that will help you and rework code or create custom plugins to do exactly what you want

For ease of use, functionality, and scaleability, Amember with e107 is probably my favorite pick.
